Within Prague, walk or use the excellent and cheap tram/metro system (buy a 24-hour or 30-minute ticket). For Day 2, the most efficient way to Český Krumlov is by direct bus (RegioJet/FlixBus). The journey takes about 3 hours each way. Trains require a change and are slower.
